چکیده

The small fourth chromosome of Drosophila melanogaster, also known as the dot chromosome, is unlike the other three chromosomes of Drosophila in that it contains largely heterochromatic sequence. The 1.2 Mb arm of the dot chromosome has normal gene density, but a higher frequency of repetitive sequences compared to the euchromatic arms of chromosomes 13. The many genes packaged in the dot chromosome’s heterochromatic regions are expressed more strongly than genes in the heterochromatic regions of other chromosomes. This unusual chromosome provides an opportunity to study differences in euchromatin and heterochromatin coding and formation. To investigate the effects of sequence organization on formation of heterochromatic and euchromatic domains, Bio 4342 has used comparative genomics, looking at dot chromosome sequences from D. melanogaster and D. virilis. 2007 marks the start of analysis of a new dot chromosome – that of D. mojavensis. My project focuses on finishing clone 430-O17 from this chromosome.

The Vivitrol pilot program (VPP): initial quantitative findings from an extended-release naltrexone study

Background In 2010, the FDA approved the use of extended-release injectable naltrexone (Vivitrol©) for the treatment of opioid dependence. Preliminary evidence from prior studies indicates that monthly injections of Vivitrol are effective in treating opioid dependence after initial detoxification. This longitudinal exploratory study measures demographics, medication adherence, sobriety (urinaly...

Cell-type Dependent Alzheimer's Disease Phenotypes: Probing the Biology of Selective Neuronal Vulnerability

Alzheimer's disease (AD) induces memory and cognitive impairment in the absence of motor and sensory deficits during its early and middle course. A major unresolved question is the basis for this selective neuronal vulnerability. Aβ, which plays a central role in AD pathogenesis, is generated throughout the brain, yet some regions outside of the limbic and cerebral cortices are relatively spare...

Comparison and Optimization of hiPSC Forebrain Cortical Differentiation Protocols

Several protocols have been developed for human induced pluripotent stem cell neuronal differentiation. We compare several methods for forebrain cortical neuronal differentiation by assessing cell morphology, immunostaining and gene expression. We evaluate embryoid aggregate vs. monolayer with dual SMAD inhibition differentiation protocols, manual vs. AggreWell aggregate formation, plating subs...

Bistable front dynamics in a contractile medium: Travelling wave fronts and cortical advection define stable zones of RhoA signaling at epithelial adherens junctions

Mechanical coherence of cell layers is essential for epithelia to function as tissue barriers and to control active tissue dynamics during morphogenesis. RhoA signaling at adherens junctions plays a key role in this process by coupling cadherin-based cell-cell adhesion together with actomyosin contractility. Here we propose and analyze a mathematical model representing core interactions involve...
